Description :
- Create and test a tool to enable a multiphoton cryo-microscope to observe phase shifts in animal germplasm under directional freezing.
- The outcome of cryopreservation varies greatly when ice crystals grow out of control during spontaneous freezing.
- To provide optimal temperature control and suitability for multiphoton microscopy, the design, manufacture, and validation of a directional freezing cryostage will be part of this process.
- LLS Rowiak will actively assist in conducting the system’s validation.
- Application of reproductive cells and the appropriate fluorescent dyes (such as Hoechst33342, CellLight Plasma Membrane-CFP) will be used to monitor the integrity of the cell’s internal structure and any membrane alterations that occur during cryopreservation.
- Create sophisticated picture processing and analysis software.
- Images taken using a multiphoton cryo-microscope will be utilized to develop software for image processing and analysis. This particular objective will be conducted in cooperation with NURE.
- Investigate samples with multiphoton cryo-microscope during freezing and thawing process including image analysis. The effect of freezing parameters (temperature gradient, CPA/VA type, cooling rate) on the integrity of reproductive cells will be analyzed.
