Description :
- To track CPA/VA diffusion inside the cells, create and validate a Raman microscopy equipment with a temperature-controlled chamber.
- It is crucial to comprehend how CPAs and VAs diffuse into cells in order to ensure effective cryopreservation. The institution’s mechanical workshop will design, build, and test the diffusion chamber using reproductive cells. Following certain RAMAN peaks of the individual components of the CPA/VA solution (DMSO, ethylene glycol, formamide, propylene glycol, sucrose, and trehalose), the diffusion of CPA/VA will be seen in a dynamic perfusion chamber.
- The calculated diffusion coefficients will be contrasted with those obtained from an examination of CPA/VA diffusion using cryomicroscopy based on variations in cell volume (NASU partner).
- Utilizing various cooling rates, CPAs/VAs, and sperm and oocyte structural changes caused by cryopreservation.
- Among the structural changes appearing during cooling/warming procedures as well as addition and removal of CPAs/VAs we will focus on analysis of cytoplasmic lipid reduction, membrane fatty acid composition alteration, cholesterol and aquaporin content.
